Rapid Strip Test for Aflatoxin M1 Residues in Milk

>The AuroFlow™ Aflatoxin M1 Strip Test Kit is a qualitative and rapid lateral flow assay designed to detect Aflatoxin M1 residues in raw commingled cow’s milk at 0.5 ppb. This immunochromatographic assay eliminates the need for a heating block. This test is designed for field use or reference lab settings

The assay uses a competitive colloidal gold based format. The milk sample (200 μL) is added to a clear plastic reaction vessel, and used to resuspend the lyophilized reagents to a uniform pink color in the bottom of the microtiter wells. The milk is incubated at room temperature to allow the antibodies on the gold particles to engage with any Aflatoxin M1 present in the milk. The test strip is then inserted into the sample well with the arrows pointing downward initiating capillary flow up the strip. Any gold particles that are not complexed with Aflatoxin M1 present in the milk will bind to the Aflatoxin M1 imprinted at the Test line (T-line), forming a signal (red line) at that position. If the antibody on the gold particle has engaged with antibiotic present in the milk sample, the gold particle will flow past the T-line and reach the Control line (C-line). For visual interpretation of the test results, T-line signal intensity that is stronger than the signal at the C-line indicates a negative result. Signal at the T-line which is equal or less intense compared to the C-line indicates the presence of 0.5 ppb or greater of Aflatoxin M1 in the milk. The greater the reduction in signal intensity at the T-line, the greater the concentration of Aflatoxin M1 present in the milk.

Aflatoxin M1 is the hydroxylated metabolite of Aflatoxin B1. Aflatoxins are believed to be the most carcinogenic naturally occurring substances. In 1987, the WHO classified the aflatoxins as Group 1 carcinogens. Aflatoxins are potent liver carcinogens and DNA-damaging agents from natural source. Aflatoxin M1 is known to be oxidized by the mixed function oxygenases of the liver P-450 system. This oxidation results in a reactive 8,9-epoxide as the major product, which seems to preferentially attack certain guanine residues in double stranded DNA. Aflatoxicosis is poisoning that results from ingestion of aflatoxins in contaminated food or feed. The LD50 of Aflatoxin M1 is 0.3 mg/kg (oral, duckling).

The AuroFlow Aflatoxin M1 Strip Test Kit has the capacity for 96 (Cat # 1101-01) or 192 (Cat # 1101-02) determinations. The shelf life is 24 months when the kit is properly stored.

The positive and negative control vials contain lyophilized raw milk samples, and must be reconstituted with 1 mL of distilled water before use. The reconstituted raw milk controls are stable for 48 hours at 4°C. Additional positive and negative control vials are available separately (Cat # 110110).
