The MaxSignal® Fructose Assay Kit is a cuvette-based enzymatic assay for the determination of fructose across a variety of food samples and fermentation media. The assay is useful for analysis of inverted sugar, a mixture of equal parts of glucose and fructose resulting from the hydrolysis of sucrose; this sugar is found naturally in fruits and honey and produced artificially for use in the food industry. The assay can also be performed using a microtiter plate for high-throughput sample analysis.
The MaxSignal® Fructose Assay Kit uses a highly-specific coupled enzymatic reaction to detect fructose in food samples: Fructose is first specifically converted to fructose-6-phosphate (F-6-P) using hexokinase. Next the F-6-P is converted to glucose-6-phosphate (G-6-P) using the enzyme phosphoglucose isomerase (PGI). The G-6-P compound is then used as a substrate by the enzyme glucose-6-phosphate dehydrogenase to convert NADP+ to NADPH. This conversion, measured at 340 nm, is proportional to the level of fructose in the sample.
The absorbance of each sample at 340 nm is measured using a spectrophotometer. The concentration of fructose in each sample is then directly determined from the change in absorbance at 340 nm within 30 minutes.
